Zone electrophoresis was originally developed at Stanford University in 1952 by George Eberle, who had previously worked with Slab Electrophoreses. It uses an electric field to drive charged molecules through a porous matrix (such as agarose) that can be placed between two electrodes: one positively-charged and one negatively-charged. The larger macromolecules progress more slowly than smaller ones because they experience greater resistance from the medium; when this happens, they become concentrated around specific zones within the solution depending on how far they have progressed in comparison to other large molecules.
Paper Electrophoresis:
Paper electrophoresis was first described in 1957 by Edward Davidson. It is a technique used to separate and purify DNA molecules, RNA fragments, proteins, peptides, and other biological macromolecules. Paper electrophoresis uses an electric field to drive charged molecules through paper soaked in polyethylene glycol (PEG) or agarose solution; the larger ones progress more slowly than smaller ones because they experience greater resistance from the medium. An advantage of this method is that it can be performed at room temperature without requiring any special equipment except for some pieces of filter paper which are inserted into a gel-containing tube with plastic pipette tips on either end.
Gel Electrophoresis:
Gel electrophoresis is a technique used to separate and purify DNA molecules, RNA fragments, proteins, peptides, and other biological macromolecules. Gel electrophoresis is one of the most important and popular techniques in biological sciences. It was first developed by Arthur Pardee at Berkeley University, USA (1953), which used a slab gel for separation, followed by Frederick Sanger who introduced capillary electrophoresis to replace slab gels.
